New gold pincer-type complexes induce caspase-dependent apoptosis in human cancer cells in vitro

Background/Aim. The use of cisplatin as a chemotherapeutic opened the door to new metal-based drug research. New complexes containing metals such platinum, palladium, ruthenium and gold have recently been analyzed potential antitumor agents. aim study was investigate cytotoxicity Au(III) with pincer-type ligands against cervical carcinoma cells (HeLa), breast cancer (MDA-MB-231 4T1) colon (HCT116 CT26), well examine type mechanism cell death that these induced in cells. Methods. investigated by MTT assay. apoptosis treated measured flow cytometry applying Annexin V/7AAD staining. expressions active proapoptotic protein Bax, antiapoptotic Bcl-2 percentage cleaved caspase-3 were determined cytometry. Results. Complex 1 showed most potent effect on HeLa cells, both compared other two examined cisplatin. IC50 values after 72 hours 1.3 ? 0.4 ?M, 3.4 5.7 0.6 26.7 6.5 ?M for 1, 2, 3 cisplatin, respectively. also exhibited highest MDA-MB-231 HCT116 tested compounds. results staining all three Our caspase-dependent mechanism, but we did not observe activation an internal pathway occurred Conclusion.
